In Vivo PEI + PSar platform
Curapath’s In Vivo platform combines linear PEI with a PEG-free shielding polymer (polySarcosine based) to form shielded polyplexes for nucleic acid delivery. This approach enables a reduced toxicity by eliminating the commonly associated immunogenity associated to PEI.

About Curapath® PEI + PSar in vivo platform
The Curapath® PEI + VLC3 In Vivo Platform combines linear polyethylenimine hydrochloride (PEI, MW 20,000 Da) with Curapath’s patented VLC3 shielding technology, based on polysarcosine. PEI complexes nucleic acids (particularly DNA) into polyplexes, while VLC3 adds a shielding layer designed to support their delivery in vivo.
By combining DNA complexation with polymer shielding, the platform aims to reduce PEI-associated toxicity and unwanted immune activation while supporting efficient nucleic acid delivery. This approach is being explored for in vivo applications, including DNA vaccine delivery and gene delivery
Potential Applications
The Curapath® PEI + VLC3 platform is designed to explore shielded nucleic acid delivery in vivo, with a particular focus on DNA. Application fit will depend on the nucleic acid, formulation, and route of administration.
DNA Vaccine Research
PEI-based systems have been investigated for plasmid DNA vaccine delivery. Curapath PEI + VLC3 can be evaluated as a shielded polyplex approach to support DNA delivery and antigen expression in vaccine research.
Localized In Vivo Gene Delivery
VLC3 can be modified with a targeting agent to direct the polyplex toward selected cells or tissues. This flexibility supports the development of targeted in vivo DNA delivery approaches, including localized gene delivery.
Curapath® PEI + VLC3 In Vivo Platform
How It Works
Curapath® linear PEI complexes DNA to form a polyplex. The VLC3 shielding polymer, based on polysarcosine, is then used to create a shielded polyplex for in vivo nucleic acid delivery.
VLC3 is a PEG-free specialty additive designed to improve nanoparticle physicochemical properties. It can also be modified with a targeting agent to direct the polyplex toward selected cells or tissues.

C5 Activation
VLC3 is a PEG-free shielding polymer designed to improve nanoparticle physicochemical properties and address a key challenge associated with PEI for in vivo delivery: unwanted immune activation.
In the data shown, shielded nanoparticles display lower C5 activation than PEI alone under the conditions tested.

Formats and supply
For R&D, four ready-to-test packs are available at different concentrations. GMP supply can be provided in custom formats to meet project requirements. The standard presentation is dry, with solution format available upon request.
Frequently Asked Questions About the PEI + PSar In Vivo Platform
What is Curapath
Curapath is a European Contract Development and Manufacturing Organization (CDMO) specialized in non-viral drug delivery systems. We support biotech and pharmaceutical companies from early development through GMP-ready and commercial manufacturing, combining proprietary delivery components, in-house technologies, and deep expertise in complex formulation and CMC development.
What is the Curapath® PEI + VLC3 In Vivo Platform?
The Curapath® PEI + VLC3 In Vivo Platform combines linear polyethylenimine hydrochloride (PEI) with VLC3, a polysarcosine-based shielding polymer. PEI complexes nucleic acids into polyplexes, while VLC3 provides a shielding layer designed to support their delivery in vivo.
How does the PEI + PSar platform work for in vivo nucleic acid delivery?
Linear PEI first complexes nucleic acids, particularly DNA, to form polyplexes. The polyplex is then combined with the VLC3 shielding polymer to create a shielded delivery system. This approach is designed to support in vivo nucleic acid delivery while improving the physicochemical properties of the polyplex.
What is VLC3?
VLC3 is Curapath’s patented polysarcosine-based, PEG-free shielding technology. It is designed to form a protective layer around PEI-based polyplexes and can be modified with a targeting agent for selected cells or tissues.
Can the platform be used for DNA vaccine research?
Yes. The PEI + VLC3 platform can be evaluated as a shielded polyplex approach for plasmid DNA vaccine research, supporting DNA delivery and antigen expression studies in vivo.
Is the PEI + VLC3 platform available in R&D and GMP formats?
For R&D, Curapath offers four ready-to-test packs at different concentrations. GMP supply can be provided in custom formats according to project requirements. The standard presentation is dry, with solution format available upon request.
How can I select the right format for my in vivo programme?
